Promoter Live Nation says Tuesday's performance at Qudos Bank Arena won't go ahead based on medical advice.
The American singer was admitted to St Vincent's Hospital in Sydney on Sunday night with dehydration but later discharged.
She was readmitted on Monday and diagnosed with a gastric virus.
"Pink remained in hospital overnight, continuing her treatment and recovery. She will be reassessed by her doctors today," Live Nation said in a statement on Tuesday.
The 38-year-old cancelled her first Sydney show on Friday night on doctor's orders, but battled through a performance on Saturday night.
